pH Acceptable pH range is 6.4-7.5 at 25 °C. Please note that the pH of SOB Medium liquid increases as the temperature of the solution decreases.
Preparation Method Reagents are tested and chosen for their ability to promote optimum growth of E. coli when combined in SOB formulations. Chosen reagents are combined in the proper proportions according to the SOB formulation and thoroughly blended for a uniform distribution. A sample of the blended formulation was used to prepare liquid medium by adding 31 g of powder per liter of purified water. The combinations were mixed by hand for about 1 minute to dissolve and autoclaved at 121°C for 15 minutes. Autoclaving at higher temperatures or for longer periods will result in darkening of the sterile medium, (a darker color does not noticeably affect growth of cells, only the appearance of the sterile medium).
