Single dropout formulation (without Leucine) of Synthetic Defined (SDA) Yeast Media with agar includes all necessary components for selective growth of Saccharomyces cerevisiae.

Application Notes

The selection of plasmids in yeast is based on the use of auxotrophic mutant strains that cannot grow without a specific media component (an amino acid, purine or pyrimidine). Transformation with a plasmid containing the mutated gene enables the transformant to grow on a medium lacking the required component. This CSM-Dropout medium create a richer medium for better yield and growth rate, and increase the probability of successful transformations when screening libraries or performing gene knock-outs.

Preparation Method Reagents are tested and chosen for their ability to promote optimum growth of yeast strains when combined in the Synthetic Defined (SD) or Synthetic Defined Agar (SDA) or SD/SDA-Dropout formula. SD is 1.7 g Yeast Nitrogen Base, 5 g Ammonium Sulfate and 20g Dextrose per liter. SDA is SD with 17% agar. Chosen reagents are combined in the proper proportions according to the formulation listed in the certificate of analysis and milled to a powder which thoroughly blends the reagents for uniform distribution. A sample of the milled and blended formulation was used to prepare liquid medium by adding 27.49 SD powder or the appropriate weight ofSD-Dropout to a liter of purified water. The combinations were mixed by hand for about 1 minute to dissolve the dextrose and autoclaved at 121 °C for 25 minutes.
Solubility Soluble in water.
Sterilization of Solutions Autoclave at 121 °C for 15 minutes.
